The immunological system remains susceptible to a number of unforseeable assaults such as cancer, surgery or to the often unavoidable administeration of therapeutic chemicals and drugs. In a double masked, placebo controlled comparative clinical trial in Corneal Ulcer and Conjunctivitis patients, DEFENSEPLUS was given orally with Ciprofloxacin eye drops 0.3% topically. Statistically significant results were seen for faster healing of corneal ulcers alongwith quicker recovery from congestion in DEFENSEPLUS treated patients. Similarly this combination therapy gave significantly better results in Conjunctivitis alongwith faster relief from itching, foreign body sensation, papillae and irritation. Statiscally significant rise was observed in the lymphocyte count of cancer patients treated with DefensePlus and they quickly recovered from Leucopaenia. In 30 patients suffering from Dengue fever (a viral infection) with profound Thrombocytopaenia (Platelet count <50000), the administration of DEFENSEPLUS in a dose of 1 capsule twice daily progressively increased their platelet count well beyond 50,000 within 48-96 hours and it returned to near normal level in 7-10 days. No hemorrhagic complication could develop in these cases, there was no need for platelet transfusion and their quick recovery was facilitated. Bacterial activity of neutrophils is an important defense mechanism. DEFENSEPLUS significantly stimulated phagocytosis and bactericidal capacity of neutrophils (%of positive cells was increased from 16.98+/- 1.23 to 23.71+/- 1.93) Significant increase in soluble immune complex (from 17.09 +/- 1.96 to 26.21 +/- 1.12) also indicates immune potentiating action of DEFENSEPLUS.
DEFENSEPLUS caused a statistically significant increase in the antibody litres in both the primary (from 4.166±0.48 to 6.166 ±0.44) and secondary immunity assay (from 5.5±0.22 to 7.0±0.26), mediated by lgC3 or IgM class of antibodies. In another study, the combined treatment with DEFENSEPLUS against Staph aureus infection reduced the mortality to 41.66% as compared to 66.66% in the antibiotic (Tetracycline) atone treated group. In another experiment in caecal ligation induced abdominal infection, the mortality in DEFENSEPLUS combined group was found to be 16.66% as compared to 41.55% in antibiotic (Metronidazole + Gentamicin) alone group. In both the experiments, 100% mortality was observed in untreated control groups. These results indicate that DEFENSEPLUS enhances the therapeutic success rate of both bacteriostatic and bactericidal antibiotics by optimising host immune status. Looking to declining success rate of antibiotics and increasing bacterial resistance, DEFENSEPLUS can be used as an adjuvant to antimicrobial therapy for better control of infections. DEFENSEPLUS significantly increased the macrophage cell size (from 36.15 ± 2.99 to 82.95 ±11.31) and maximum increase in the number of cells was found to be on the 10th day of its administration (from 1700 ± 118 to 6133 ± 663).
